Conclusions What practical relevance does this have? Manipulated or artificial chromosomes have been produced in several laboratories with the aim, at least in part, of providing an expression platform for genes that avoids both integration into host-cell genomes and position effects. Paradoxically, a potential problemwith this approach is that genes placed in close proximity to a centromere might be silenced by centromeric heterochromatin, but this now seems less of a concern. A better understanding of the requirements of a centromere in terms of formation, propagation and function in cell division is needed to produce better artificial chromosomes. The approaches discussed here that are used to define molecular markers of centromeric DNA, coupled with biochemistry and biophysics of centromeric chromatin, are a part of that process.

cis-Acting determinants of heterochromatin formation on Drosophila melanogaster chromosome four.
The heterochromatic domains of Drosophila melanogaster (pericentric heterochromatin, telomeres, and the fourth chromosome) are characterized by histone hypoacetylation, high levels of histone H3 methylated on lysine 9 (H3-mK9), and association with heterochromatin protein 1 (HP1). While the specific interaction of HP1 with both H3-mK9 and histone methyltransferases suggests a mechanism for the ...
متن کاملInfluence of DNA methylation on positioning and DNA flexibility of nucleosomes with pericentric satellite DNA
DNA methylation occurs on CpG sites and is important to form pericentric heterochromatin domains. The satellite 2 sequence, containing seven CpG sites, is located in the pericentric region of human chromosome 1 and is highly methylated in normal cells. In contrast, the satellite 2 region is reportedly hypomethylated in cancer cells, suggesting that the methylation status may affect the chromati...
متن کاملA Distinct type of heterochromatin within Drosophila melanogaster chromosome 4.
Studies of transcriptional gene silencing in Drosophila melanogaster suggest that most of chromosome 4 resembles pericentric heterochromatin. However, some modifiers of position-effect variegation, including chromosome 4 dosage and loss of SU(VAR)3-9, have different effects on silencing in pericentric vs. distal arm chromosome 4 heterochromatin, distinguishing these two heterochromatin types.
متن کاملElimination of a specific histone H3K14 acetyltransferase complex bypasses the RNAi pathway to regulate pericentric heterochromatin functions.
In Schizosaccharomyces pombe, the RNAi pathway is required for the formation of pericentric heterochromatin, proper chromosome segregation, and repression of pericentric meiotic recombination. Here we demonstrate that, when the activity of the histone H3 Lys 14 (H3K14) acetyltransferase Mst2 is eliminated, the RNAi machinery is no longer required for pericentric heterochromatin functions. We fu...
متن کاملElement 1360 and RNAi Components Contribute to HP1-Dependent Silencing of a Pericentric Reporter
In eukaryotes, distinct regions of the genome are packaged as euchromatin (less condensed, more active) or heterochromatin (condensed, silenced). Studies in yeast, plants, and flies suggest that RNA interference (RNAi) is linked to heterochromatin formation and transcriptional silencing of transposable element (TE) sequences. We previously reported that insertion of a mobile hsp70-white reporte...
